Enclomiphene is not a peptide. It is a selective oestrogen receptor modulator — a small molecule, the trans-isomer of clomiphene, taken orally. It contains no amino acids.

It sits alongside peptides in the same vendor catalogues and forums, which is how it enters these conversations. Its mechanism is worth understanding precisely because it is the opposite of what most compounds here do.

Not approved in the US, EU, or UK for this use. Enclomiphene has been the subject of clinical development and professional position statements but is not an approved treatment in these jurisdictions. Hypogonadism is a medical diagnosis requiring clinical assessment.

How it works — by removing a brake

Testosterone production is regulated by feedback. The hypothalamus releases GnRH, prompting the pituitary to release LH and FSH, which act on the testes. Testosterone and oestrogen then feed back to slow that signal.

Enclomiphene blocks oestrogen receptors in the hypothalamus and pituitary. With the feedback signal interrupted, the brain reads oestrogen as low, so GnRH, LH and FSH rise — and testosterone rises with them.

This is the crucial contrast with testosterone therapy. Supplying testosterone externally suppresses the axis, because the feedback loop still works and reads levels as adequate. Enclomiphene stimulates the axis instead, which is why it preserves the downstream signalling that exogenous testosterone shuts down.
