hot take: the amylin analogs are more interesting than a fourth receptor

amylin_nextAnalyst

Amylin is co-secreted with insulin and has its own role in satiety and gastric emptying. Analogs are being developed alone and combined with GLP-1 agents.

Why I think it's underrated: the mechanism is complementary rather than overlapping. Two agents hitting the same pathway harder gets you diminishing returns and worse tolerability. Agents on genuinely different pathways is where combination pharmacology usually pays off.

All investigational, some phase 2 combination data published, nothing approved.

I'd expect this to be more interesting than bolting a fourth receptor onto an agonist. Opinion not prediction. Change my mind.

Broadly agree, complementary pathways is sound logic. Counterpoint: combination products carry combination tolerability, and this class already has a ceiling there.

Strongest objection and I don't have a good answer. If the limit is GI tolerability, more mechanisms may not help.
