Plecanatide has an FDA-approved human dosage. Trulance for adults with chronic idiopathic constipation or IBS with constipation is 3 mg by mouth once daily, with or without food.

Where sources disagree
- Trulance is approved only for adults. Its boxed warning makes it contraindicated under 6 years of age, after deaths from dehydration in young juvenile mice, and says to avoid it from 6 to under 18.
- The label notes plecanatide is minimally absorbed: plasma levels after a 3 mg dose were mostly below the limit of quantitation, so no half-life could be calculated.
