DSIP stands for delta sleep-inducing peptide, and the name is the problem. It is a nine-amino-acid peptide isolated in the 1970s from the blood of rabbits induced into a sleep-like state, and named for the effect its discoverers were looking for.

Nearly fifty years later, the evidence that it induces sleep in humans is considerably weaker than the name implies.

Not an approved drug. DSIP has no approved human indication and no established human dose. It is sold for laboratory research only, and nothing here is dosing guidance.

Where the name came from

The original work isolated a substance from cerebral venous blood of rabbits in an induced sleep state, and reported that transferring it produced delta-wave sleep in recipients. The peptide was characterised and named for that observation.

Naming a molecule after a hoped-for function is common in early research and it shapes everything that follows. Once a compound is called "sleep-inducing peptide", every subsequent discussion starts from the assumption that it induces sleep.

This is worth watching for generally. MOTS-c gets described as an "exercise mimetic" and AOD-9604 as a "fat-loss fragment" — names that assert a conclusion the evidence has not reached.

What five decades produced

  • Sleep effects. Findings have been inconsistent, and DSIP is not established as a reliable sleep-inducing agent in humans.

  • Other reported effects. Work has described effects on stress response, pain perception, and hormone regulation — a scattered profile rather than a focused one.

  • Endogenous role. It occurs naturally, but its physiological function is not clearly established.

  • Recent work. Research continues, including on delivery approaches and fusion peptides in rodent insomnia models.

The honest position: a real molecule with a real if inconsistent literature, whose most memorable feature is a name promising more than the data support.

The blood-brain barrier question

For a peptide expected to act on sleep architecture, reaching the brain matters. Peptides generally cross the blood-brain barrier poorly, and the recent literature includes work specifically on delivery strategies and fusion constructs to address that.

That the delivery problem is still being worked on is itself informative about how settled the basic effect is.

Storage
