what does half-life actually mean in practice? not the textbook definition, the practical one

i know the definition. time for the concentration to drop by half. thats not what im asking.

what i dont get is what it actually tells you. people quote half-lives like theyre the most important property of a compound and im not sure what im supposed to do with the number.

like if something has a 30 minute half life vs 6 days, obviously thats a big difference, but a big difference in what exactly?

    Honest answer: it tells you about accumulation and dosing interval, not duration of effect. Those get conflated constantly.

    1. Steady state after roughly 4–5 half-lives. Six-day half-life means weeks before levels stop climbing, so early observations aren't representative. 2. Short half-life relative to the interval gives peaks and troughs. Long gives a flat line. 3. Washout is the same arithmetic backwards.

    What it does not tell you is how long an effect lasts. That depends on what happens downstream, which can outlast the compound entirely.

      ok the accumulation bit is what i was missing. so a long half life means week one isnt telling you much

      Exactly. It's why people conclude something "started working" when what changed was levels finally plateauing.

    Some things bind essentially irreversibly, so the effect tracks protein turnover rather than plasma levels. Half-life is nearly irrelevant to duration there.
